Abstract | La Gliconanotecnología, desarrollada en nuestro laboratorio, aborda la síntesis de nuevas nanopartículas biofuncionales con aplicaciones en biomedicina. La Gliconanotecnología, desarrollada en nuestro laboratorio, aborda la síntesis de nuevas nanopartículas biofuncionales con aplicaciones en biomedicina. La presente Tesis Doctoral presenta una aplicación de nanopartículas como inhibidores de la adhesión del Virus de Inmunodeficiencia Humana (VIH). En el presente trabajo se han desarrollado nuevas familias de Gliconanopartículas de oro, funcionarizadas con oligosacáridos de tipo "high mannose", que mimetizan la envoltura de carbohidratos del VIH y están diseñadas como inhibidores de la interacción entre el receptor DC-SIGN y la glicoproteína gp120, que constituye una etapa clave en la transmisión del VIH por vía sexual. Nuestro diseño y construcción se han validado mediante experimentos de inhibición de la interacción DC-SIGN/gp120 mediante el uso de biosensores, confirmando la afinidad de las mismas por la proteína DC-SIGN. Con estos sistemas se han realizado experimentos biológicos de inhibición de la trans-infección, de citotoxicidad y de respuesta celular. Los experimentos in vitro, utilizando un modelo que simula las condiciones naturales de la trans-infección del VIH mediante el uso de virus recombinantes y células dendríticas de humano. Los resultados obtenidos confirman nuestras nanopartículas como una buena herramienta en la inhibición de la trans-infección del VIH y abre nuevas perspectivas en el desarrollo de nuevas herramientas terapéuticas de tipo microbicida para luchar contra la infección por VIH. |
