Abstract | El presente trabajo de fin de grado se centra en el estudio de las ciclodextrinas, oligosacáridos cíclicos constituidos por unidades de glucosa unidas por enlaces α-(1,4), con estructura toroidal, capaces de formar complejos ... El presente trabajo de fin de grado se centra en el estudio de las ciclodextrinas, oligosacáridos cíclicos constituidos por unidades de glucosa unidas por enlaces α-(1,4), con estructura toroidal, capaces de formar complejos de inclusión. La industria farmacéutica utiliza tanto las ciclodextrinas naturales como sus diferentes derivados como recurso tecnológico para la mejora de la solubilidad y velocidad de disolución de fármacos poco solubles, aumentando así su biodisponibilidad oral. En el presente trabajo se recogen los requisitos necesarios y los diferentes métodos existentes para obtención de dichos complejos. Además, se describen las distintas aplicaciones que ofrecen estos excipientes en el campo farmacéutico según la vía de administración. La última parte del trabajo se centra en evaluar la producción científica en este tema en los últimos 20 años, tratando de identificar los medicamentos que se han comercializado utilizando ciclodextrinas en su formulación, tanto a nivel de la agencia española de medicamentos y productos sanitarios (AEMPS) como a nivel mundial. Sin embargo, a pesar de sus propiedades solubilizantes y del resto de ventajas que como excipiente puede aportar su utilización en una formulación, respaldada por las numerosas publicaciones al respecto, hay muy pocos medicamentos que han sido comercializados. También es frecuente encontrarnos medicamentos que fueron comercializados en su día y que posteriormente han sido retirados del mercado. A nuestro juicio, han sido factores farmacoeconómicos los responsables de esta situación ya que, aunque se trate de excipientes capaces de mejorar algunas propiedades biofarmacéuticas siguen siendo un grupo de excipientes excesivamente caros. Sólo cuando otras alternativas no funcionan, se recurre a ellos para solucionar la problemática en cuestión. |
