Abstract | El cáncer sigue siendo una de las principales causas de mortalidad a nivel mundial, detrás de las enfermedades cardiovasculares. El cáncer sigue siendo una de las principales causas de mortalidad a nivel mundial, detrás de las enfermedades cardiovasculares. Factores genéticos y epigenéticos están asociados al riesgo de contraer la enfermedad. La radioterapia y la cirujía son los tratamientos más eficaces contra el cáncer local, y la quimioterapia, terapias hormonales y biológicas frente a tumores metastásicos, capaces de alcanzar el resto de los órganos y tejidos a través del sistema circulatorio. La quimioterapia inhibe el crecimiento de las células cancerosas, pero también afecta a las células sanas. A pesar de los tratamientos tradicionales, el pronóstico de los pacientes no ha mejorado significativamente. La toxicidad y destrucción inespecífica de células normales requería la necesidad de desarrollar tratamientos alternativos dirigidos y eficientes, siendo la terapia génica una de las técnicas más prometedoras. Recientes avances en terapia génica han mejorado la tasa de supervivencia y la esperanza de vida de los pacientes, así como las posibilidades de tratamiento. Las técnicas más avanzadas y utilizadas actualmente son la terapia de células CAR-T, la viroterapia oncolítica y la técnica de edición de genes CRISPR. En este trabajo se detallan los avances sobre eficacia en terapia génica tras la realización de varios ensayos clínicos, así como los obstáculos y posibles soluciones futuras de las principales técnicas utilizadas actualmente para el cáncer. La terapia génica es una alternativa antineoplásica, que, si bien aún no es la cura para dicha enfermedad, promete obtener resultados alentadores para llegar a formar parte del arsenal terapéutico habitual. |
