granulacion de origen sinovial (pannus), ... Introducción: La artritis reumatoide (AR) es una enfermedad inflamatoria sistemica cronica de etiologia autoinmune caracterizada por una sinovitis erosiva en la cual el tejido de granulacion de origen sinovial (pannus), invade y erosiona el cartilago y el hueso de las articulaciones diartrodiales. Los fibrobalstos sinoviales (FS) o sinoviocitos de tipo fibroblastico juegan un papel fundamental en la persistencia de la inflamacion cronica y en el dano articular, siendo unas de las poblaciones celulares mas abundantes del pannus. La oleuropeina (OLE) es un secoiridoide, componente fenolico mayoritario de la pulpa del fruto y hojas de Olea europaea L. y, que a su vez, se encuentra en el aceite de oliva. Se han documentado diversas actividades farmacologicas del citado secoiridoide, entre las que se destaca la antioxidante y antiinflamatoria. No obstante no existen, hasta la fecha, estudios experimentales en fibroblastos humanos que validen su potencial antirreumatico, asi como los mecanismos bioquimicos y moleculares implicados. Objetivos: Este estudio pretende evaluar el efecto antiinflamatorio de OLE en la linea celular SW982 inducida con IL-1β, mediante el analisis de la produccion de la produccion de mediadores pro-inflamatorios, asi como, las vias de senalizacion involucradas. Materiales y métodos: Las celulas SW982 fueron cultivadas en DMEM y estimuladas con IL-1β (5 ng/mL) en presencia o ausencia de OLE (50 y 100 μM). La viabilidad celular se determino mediante un ensayo con sulforrodamina B (SRB). Los niveles de TNF-α e IL-6 fueron evaluados por la tecnica ELISA. La expresion de las proteinas inflamatorias COX-2 y m- PGES-1 y, estudio de las vias de senalizacion celular fueron llevados a cabo mediante Western blot. Resultados: El tratamiento con OLE redujo significativamente los niveles de IL-6 y TNF- α, y ademas disminuyo la sobreexpresion de COX-2 y m-PGES-1. Este compuesto a las concentraciones ensayadas fue capaz, ademas, de modular la activacion de JNK y p38, impidiendo su fosforilacion. Conclusión: OLE reduce la produccion de citocinas pro-inflamatorias IL-6 y TNF-α y la expresion proteica de COX-2 y m-PGES-1, posiblemente a traves de la inhibicion de la via de senalizacion de MAPK: PJNK y p38, en fibroblastos sinoviales humanos SW982 estimulados con IL-1β |
