enfermedad de Alzheimer (EA) y la demencia vascular, ya que tienen efectos tanto centrales
como periféricos sobre la acumulación ... Las lipoproteínas de alta densidad (HDL) juegan un papel importante en la patología de la enfermedad de Alzheimer (EA) y la demencia vascular, ya que tienen efectos tanto centrales como periféricos sobre la acumulación cerebral de beta-amiloide (Aꞵ) y las funciones vasculares. Sin embargo, sigue sin estar claro hasta qué punto las HDL pueden intercambiar sus componentes proteicos y lipídicos entre el sistema nervioso central (SNC) y la circulación sistémica. Este estudio pretende evaluar el efecto de HDL aisladas de voluntarios sanos (hHDL) y pacientes con síndrome metabólico (msHDL) en células endoteliales de microvasculatura cerebral humana (HEBC-5i), componentes principales de la barrera hematoencefálica (BHE). Las células HBEC-5i se pre-estimularon con Aꞵ (500 nM) y 1 h después se trataron con hHDL o msHDL a 300 µg de proteína/mL durante 24 h. Además, se utilizaron ApoE-e2 y ApoE-e4 como control a 10 µg/mL. Se analizó la capacidad de eflujo de colesterol y se aisló ARN para realizar RT-qPCR. Se midió la expresión de genes implicados en la respuesta inflamatoria, como IL-6, IL-1ꞵ, TNFα y transportadores ABC (ABCA1 y ABCB1). Además, se midió la expresión de la proteína relacionada con el receptor de LDL 1 (LRP1), genes del metabolismo lipídico (SR-BI, LDLR, EL, CD36, FATP y S1PR1) y moléculas de adhesión celular (CAM) como la molécula de adhesión celular vascular-1 (VCAM-1) y la molécula de adhesión intercelular-1 (ICAM-1). Nuestros resultados mostraron que las msHDL aumentaban la expresión de citocinas proinflamatorias y reducían los genes relacionados con la eliminación de Aꞵ, como ABCA1 y LRP1, en las células HBEC-5i, confirmando así que las alteraciones metabólicas asociadas a la obesidad invierten las propiedades protectoras de las HDL en la BHE. |
