Universidad de Sevilla, Sevilla.https://hdl.handle.net/11441/175995In this PhD Thesis, we have evaluated the qualitative and quantitative changes in the HIV-1 reservoir in lymphocytes T CD4+, the phenotypic changes in lymphocytes T CD4+ and CD8+, different inflammatory markers, and immune recovery after 12 and 24 months of starting a dual antiretroviral therapy (DT) based on dolutegravir/lamivudine (DTG/3TC) compared with a triple therapy (TT) based on dolutegravir + tenofovir alafenamide/emtricitabine We also compared the viral kinetics in seminal plasma in the same clinical trial between these two treatment arms. Finally, we analyzed the long-term impact on the HIV-1 viral reservoir, immunological parameters, various inflammatory markers, and immune recovery after simplification to DT based on DTG/3TC or darunavir cobicistat + lamivudine (DRVc + 3TC) for ?4 years compared to maintaining a triple therapy (TT) based on bictegravir, tenofovir alafenamide, and emtricitabine (BIC/TAF/FTC). In the three studies, we have shown no differences in any of the parameters analyzed, which clarifies previous doubts about the possible consequences of using these DTs instead of a standard three-drug antiretroviral treatment.
