Abstract | En nuestro estudio hemos tratado de profundizar en la etiopatogenia de la enfermedad ulcerosa desarrollada por ibuprofeno (ib) y diclofenaco y(dc). Los objetivos propuestos son: 1.-evaluar el daño mediado por estos sobre ... En nuestro estudio hemos tratado de profundizar en la etiopatogenia de la enfermedad ulcerosa desarrollada por ibuprofeno (ib) y diclofenaco y(dc). Los objetivos propuestos son: 1.-evaluar el daño mediado por estos sobre la mucosa gastrica de rata. 2.-estudiar los mecanismos implicados en la gastrolesividad, valorando fundamentalmente la importancia del componente vascular en el origen de las lesiones. Para ello se han analizado los posibles cambios de permeabilidad capilar, infiltrado leucocitario, implicacion de los radicales libres y papel del oxido nitrico. Ib y dc provocan lesiones que aumentan con la dosis, nuestros datos indican que los neutrofilos estarian involucrados pero cuando las dosis son muy altas estos leucocitos parecen perder su protagonismo en el daño. Hemos determinado la concentracion mucosal de compuestos sulfridrilicos totales y no proteicos, asi como, la actividad de la enzima glutation peroxidasa. Segun los resultados obtenidos podriamos hablar de mecanismos antioxidativos desencadenados tras la administracion de dc, aunque distintos a los estimulados tras el tratamiento con ib. Los estudios sobre permeabilidad vascular mucosal sugieren que las lesiones provocadas por estos dos aines no estan relacionadas con cambios de esta. Los resulados obtenidos del estudio microscopico confirman los observados macroscopicamente. Ni ib ni dc modifican la relacion mastocitos desgranulados/mastocitos totales, lo que indica que no producen cambios en la funcion mastrocitaria. Nuestros resultados sugieren un claro efecto protector de arginina independiente de su esteoisomeria. El mecanismo implicado no estaria totalmente relacionado con la presencia de oxido nitrico en la mucosa. Sin embargo, cuando se inhibe su sintesis, se observa un empeoramiento del daño. |
